Biochem Physiol Actions: Syntaxin-binding protein 1 (STXBP1) is required for secretion of neurotransmitters and for vesicle fusion in eukaryotic cells. In neurons, Mint 1 and 2 proteins bind to STXBP1 to form the essential synaptic vesicle fusion machinery and help in regulated exocytosis. It also forms a tight complex with the Soluble NSF attachment protein receptor (SNARE) syntaxin 1, in which syntaxin is held in a closed conformation and further stabilized to form the SNARE complexes. This shows that STXBP1 allows the formation of a complex between syntaxin and Synaptosomal-Associated Protein 25kDa (SNAP-25), which in turn functions as an acceptor for vesicle-bound synaptobrevin. All these activities are essential intermediates in the pathway towards exocytosis.
