Biochem Physiol Actions: The TALE (three amino acid loop extension) superfamily of proteins are involved in the regulation of transcription. This protein stabilizes the homeoprotein-DNA complex by binding to HOX or PBX proteins to form dimers. It also binds to DNA-bound dimer of PDX and HOX proteins. It associates with Pbx1, a homeodomain protein, and serves as a transcriptional regulator of KLF4, a Kruppel-like factor that has been implicated in tumorigenesis and maintenance of stem cell pluripotency. This gene may be involved in palate closure and its haploinsufficiency has been linked to orofacial clefting and learning disability.
