Biochem Physiol Actions: Cytochrome c oxidase (COX) is the terminal enzyme of the respiratory chain having subunits originating both from the mitochondrial and nuclear genome. Deficiency of this gene causes a MELAS-like syndrome. Its biogenesis defect does not involve a primary alteration in the expression of the COX subunits. Its expression is translationally regulated as in most bona fide COX assembly mutants. It is found that assembled ATPase and its proton pumping function are essential for COX biogenesis in yeast and mammalian cells through a mechanism independent of Cox1p synthesis.
